Validation and Quality Control of Polymerase Chain Reaction Methods Used for the Diagnosis of Infectious Diseases

چکیده
The most common direct detection methods are isolation or in-vitro cultivation, electron microscopy, immunofluorescence, immunohistochemistry, antigen enzyme-linked immunosorbent assay (antigen-ELISA), nucleic-acid hybridisation (NAH), macroand microarrays and the various techniques of nucleic acid amplification, such as the polymerase chain reaction (PCR) or the isothermal amplification methods, such as nucleic acid sequence based amplification (NASBA), Invader or loop-mediated isothermal amplification (LAMP). As NAH, macroand microarrays and the various amplification assays have nucleic acid molecules as targets, they are also termed methods of molecular diagnosis.

Comparison of Quantitative Real-Time Polymerase Chain Reaction (qRT-PCR) and Agglutination Assays in Diagnosis of Brucellosis in Golestan Province, North of Iran
Introduction: Brucellosis is one of the most common zoonotic infections worldwide. The clinical symptoms of brucellosis are similar to a wide range of diseases; hence, reliable diagnostic and laboratory methods are required to identify the causative agent. Iran is an endemic region of brucellosis, and many patients are misdiagnosed due to the nature of the infection. In this study, we aimed to ...
متن کاملDetection Of Toxoplasma Gondii and Human Cytomegalovirus DNA in Blood from Transplant Recipients Using Multiplex Nested Polymerase Chain Reaction
Evidences from many studies suggested a polymerase chain reaction (PCR) as a valuable method for diagnosing infectious disease in the transplant recipients. We used this method for detection of Toxoplasma, gondii and human cytomegalovirus in blood specimens from patients after bone marrow or kidney transplantation. DNA of both infectious agents were detected using two separate sets of nested pr...
متن کاملDiagnosis of bacteremia in patients with suspected septicemia using polymerase chain reaction method
Background: Sepsis or blood stream infection is a clinical lethal syndrome with severe systemic inflammatory response to infection, if not treated quickly, is associated with dangerous consequences and high morbidity and mortality. The traditional and conventional method for identification of sepsis is blood culture method which is so time-consuming and long that it eliminates the possibility o...
متن کاملMolecular analysis of the S1 gene of vaccine strains of infectious bronchitis virus using reverse transcriptase-polymerase chain reaction and restriction fragment length polymorphism
Infectious bronchitis virus (IBV) is an acute and contagious viral disease of poultry that affects different systems, including the respiratory tract in particular. IBV causes major economic losses in the poultry industry globally. Due to antigenic variation of the causative agent, control of the disease is difficult. To control the disease, many vaccines that belong to different serotypes are...
متن کاملDetection of Nocardia Asteroides Complex in Clinical Isolates by Real-Time Polymerase Chain Reaction
Background and Aims: Nocardia asteroides complex is the most common cause of infectious diseases due to nocardiosis. Interspecies differentiation of Nocardia genera is essential for prognosis and timely proper treatment, as well as for epidemiological studies. Since each genus has its own antibiotic resistance, precise careful diagnosis is of prime importance. As compared to biochemical and phe...
